"The genetics of Alzheimer's disease in the post-genomic era: towards a better understanding of the pathophysiological processes?"

Advances in high-throughput genomic approaches have significantly advanced research into the genetics of Alzheimer’s disease in recent years. To date, more than 91 loci associated with an increased risk of developing the disease have been identified. However, this knowledge is only meaningful if we understand how these genetic factors contribute to the disease's pathophysiological processes. Alongside conducting large-scale studies to further characterise the missing genetic component of AD, we are developing research projects on other neurodegenerative diseases, such as vascular dementia, to determine whether neurodegenerative diseases overlap in any way, which could help us to identify relevant pathophysiological pathways.  We are also developing induced pluripotent stem cells models edited for variants of interest to propose new pathophysiological hypotheses, particularly to go beyond the amyloid cascade. This led us to propose a genetically driven synaptic failure hypothesis.
