A technique for making organs and tissues optically transparent in order to facilitate their observation under the microscope.
This technique makes it possible to visualize a complex environment (e.g. the brain) in its entirety without resorting to serial cuts.
The Disco method is an example of a transparisation technique that has the advantage of preserving the morphology and size of the samples and thus obtaining a better resolution for image acquisition.
Principle:
- Step 1: Bleaching and permeabilization of samples
- Step 2: Immunolabelling
- Step 3: Chemical transcription to remove lipids
- Step 4: Impregnation in the mounting medium
